Overview

NI-2101 (FcRH5xCD28 κλ body) is a bispecific antibody designed to provide conditional T-cell co-stimulation for the treatment of multiple myeloma. Developed by Light Chain Bioscience using their proprietary κλ-body platform, the molecule simultaneously targets Fc receptor-like 5 (FcRH5), which is highly expressed on malignant plasma cells, and CD28, a key co-stimulatory receptor on T cells. NI-2101 is engineered as a "Signal 2" bispecific, meaning it triggers CD28 signaling only upon binding to FcRH5-positive tumor cells. This conditional activation is intended to enhance the efficacy, proliferation, and persistence of T cells when used in combination with "Signal 1" therapies, such as CD3-targeting bispecific antibodies (e.g., cevostamab) or CAR-T cell therapies, while minimizing the systemic toxicity and cytokine release syndrome typically associated with non-conditional CD28 agonism.
