FcRL5 CAR-T is a chimeric antigen receptor T-cell (CAR-T) therapy engineered to target cells expressing Fc receptor-like 5 (FcRL5), a protein highly upregulated on the plasma cells of multiple myeloma. The construct typically utilizes an anti-FcRL5 single-chain variable fragment (scFv) linked to a co-stimulatory domain (commonly CD28 or 4-1BB) with a CD3 signaling domain. Some versions are engineered to express interleukin-15 (IL-15) to enhance T-cell persistence and central memory phenotype, improving cytotoxicity and anti-tumor activity. FcRL5 CAR-T is being developed as a potential immunotherapy for patients with multiple myeloma, especially those with poor prognosis features such as gain of chromosome 1q21[1][2][3][4].
