Overview

FCRL5 CAR T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target Fc receptor-like 5 (FCRL5), a surface protein highly and specifically expressed on malignant plasma cells in multiple myeloma (MM) and certain B-cell lineages. FCRL5 is a member of the immunoglobulin receptor superfamily and is considered a promising therapeutic target because its expression is largely restricted to the B-cell lineage, particularly mature B cells and plasma cells, with minimal expression on other healthy tissues. The therapy involves engineering a patient's own T cells to express a CAR that recognizes FCRL5, enabling the selective identification and elimination of myeloma cells and potentially myeloma-initiating cells (MICs). Clinical studies in patients with relapsed or refractory multiple myeloma (RRMM) have demonstrated that FCRL5-targeted CAR-T cells can achieve promising safety profiles and clinical efficacy, particularly in cases where 1q21 amplification (1qAmp) drives disease progression.
