Overview

FCRL5 CAR-T clone 942 is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target Fc receptor-like 5 (FCRL5), a protein specifically expressed on the surface of malignant plasma cells in multiple myeloma. The therapy utilizes a humanized single-chain variable fragment (scFv) derived from clone 942, which specifically binds to the membrane-proximal D9 domain of the FCRL5 extracellular region. This binding site is strategically chosen to enhance the formation of the immunological synapse. The CAR construct incorporates a 4-1BB costimulatory domain and a CD3z signaling domain to drive T-cell activation and persistence. Preclinical evaluations in mouse xenograft models have demonstrated that clone 942 significantly reduces tumor burden and extends survival, supporting its potential as a therapeutic candidate for relapsed or refractory multiple myeloma.
