Overview

FCRL5 CAR-T clone 947 is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target Fc receptor-like 5 (FCRL5), a cell surface marker highly expressed in multiple myeloma (MM) with restricted expression on normal tissues. Clone 947 was developed using humanized single-chain variable fragment (scFv) sequences obtained from ATX mice (Alloy Therapeutics) and specifically targets the membrane-proximal D9 domain of FCRL5 to enhance the formation of the immunological synapse. The CAR construct incorporates a 4-1BB costimulatory domain and a CD3ζ signaling domain. Preclinical studies have demonstrated that clone 947 exhibits potent cytolytic activity against FCRL5-expressing myeloma cell lines and provides significant survival benefits in mouse xenograft models of aggressive MM. It is currently being positioned as a clinical candidate for the treatment of relapsed or refractory multiple myeloma.
