Overview

FCRL5 CAR-T clone 977 is an experimental chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of multiple myeloma (MM). It targets Fc receptor-like 5 (FCRL5), a cell surface protein universally expressed on primary MM cells with restricted expression on normal tissues. Clone 977 was derived from humanized antibodies generated using Alloy Therapeutics' ATX mice and specifically targets the D1-8 extracellular domains of FCRL5. The CAR construct incorporates a 4-1BB costimulatory domain and a CD3z signaling domain. Preclinical in vitro and in vivo studies have demonstrated that clone 977 possesses potent cytolytic activity against myeloma cell lines and significantly improves survival in mouse models. Additionally, clone 977 has been utilized in the development of bi-targeted CARs, such as those co-targeting BCMA or multiple FCRL5 epitopes, to enhance efficacy and prevent antigen escape.
