FCRL5 CAR-T clone 980 is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target multiple myeloma. It specifically recognizes the D9 domain, a membrane-proximal region of the Fc receptor-like 5 (FCRL5) protein, which is universally expressed on myeloma cells but has limited expression on healthy tissues. The CAR construct incorporates a 4-1BB costimulatory domain and a CD3zeta signaling domain to enhance T-cell persistence and activation. Preclinical data demonstrate that clone 980 effectively kills FCRL5-expressing myeloma cells in vitro and significantly reduces tumor burden while extending survival in mouse xenograft models. It is also being explored in bi-targeted CAR-T configurations, such as FCRL5-FCRL5 or BCMA-FCRL5 constructs, to mitigate antigen escape and enhance therapeutic efficacy.
