FD-004 is a preclinical-stage degrader-antibody conjugate (DAC) targeting Human Epidermal Growth Factor Receptor 2 (HER2), currently under development by Chengdu FenDi Pharmaceutical. The drug is built upon the company's proprietary PRODED™ technology platform, which focuses on targeted protein degradation (TPD). FD-004 is designed to utilize the high specificity of a monoclonal antibody to deliver a degrader payload directly to HER2-positive cancer cells. Once internalized, the degrader payload facilitates the ubiquitination and subsequent proteasomal degradation of the HER2 protein. This mechanism of action aims to provide a more robust therapeutic effect compared to traditional HER2 inhibitors or antibody-drug conjugates (ADCs) by completely removing the target protein and potentially overcoming resistance associated with HER2 overexpression or mutations. As of early 2024, FD-004 remains in the discovery and preclinical optimization phase.
