fd-HH phage is a genetically engineered filamentous phage (fd phage) developed as a tumor-targeting immunotherapeutic bionanofibre. It is designed to display two distinct peptides: one that binds to the extracellular domain of Programmed cell death-ligand 1 (PD-L1) and another that targets melanoma cells and tissues. Upon intravenous administration, the phage homes to the tumor site and binds to PD-L1 on cancer cells, effectively blocking the PD-1/PD-L1 signaling pathway. This blockade restores the anti-tumor activity of T cells while minimizing systemic toxicity by concentrating the therapeutic effect within the tumor microenvironment. The construct was developed by researchers at Sun Yat-sen University and has demonstrated efficacy in preclinical melanoma models.
