Drug intelligence / Profile preview
FDW028 is a potent and highly selective small-molecule inhibitor of fucosyltransferase 8 (FUT8), an enzyme responsible for the core fucosylation of N-glycans. Developed by researchers at Soochow University, FDW028 binds to the GDP-fucose binding pocket of FUT8, preventing the core fucosylation of key glycoproteins, most notably the immune checkpoint molecule CD276 (B7-H3). Defucosylation of CD276 destabilizes the protein and triggers its degradation via the chaperone-mediated autophagy (CMA) pathway. In preclinical models, FDW028 has demonstrated significant anti-tumor activity, suppressing tumor growth and metastasis in colorectal cancer and muscle-invasive upper tract urothelial carcinoma (MI-UTUC).
