Fel d 1 chain 1-derived peptides are synthetic peptides based on the T-cell epitopes of the major cat allergen Fel d 1, specifically from its chain 1 sequence. These peptides are designed to induce immune tolerance in individuals with cat allergy by modulating T-cell responses without triggering IgE-mediated allergic reactions. The most clinically studied formulation is known as Cat-PAD, which consists of a set of short synthetic peptides derived from the primary structure of Fel d 1. Administered intradermally over a short course (typically four doses over three months), these peptides have demonstrated persistent reduction in rhinoconjunctivitis symptoms for up to two years after treatment in cat-al"lergic subjects following standardized allergen challenge[2][1]. The mechanism involves induction of regulatory T cells and immune deviation away from Th2-type allergic responses[5]. This approach represents a new class called Synthetic Peptide Immuno-Regulatory Epitopes (SPIREs).
