This regimen combines fentanyl, a potent synthetic mu-opioid receptor agonist that provides rapid systemic analgesia, with ketorolac, a non-steroidal anti-inflammatory drug (NSAID) that inhibits cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) enzymes to reduce prostaglandin synthesis. In a Phase 4 clinical trial conducted by the University of South Alabama, this intramuscular combination is used as the active comparator to evaluate the efficacy and opioid-sparing potential of the Nerve of Arnold block regional anesthesia technique. The combination aims to provide effective pain control while balancing the side effects associated with opioid use in children.
