The combination of fentanyl and propofol is widely used for procedural sedation and anesthesia in various clinical settings, including outpatient procedures, endoscopy, cataract surgery, and minor surgical interventions. Fentanyl is a potent synthetic opioid analgesic that acts primarily as a mu-opioid receptor agonist to provide pain relief. Propofol is an intravenous anesthetic agent that acts as a positive allosteric modulator of the gamma-aminobutyric acid type A (GABA-A) receptor to induce hypnosis and sedation[6][5]. When used together, these drugs have synergistic effects—fentanyl enhances analgesia while allowing lower doses of propofol for effective sedation. This combination results in rapid onset of action, stable hemodynamic profiles with decreased heart rate and blood pressure, reduced drug requirements for both agents, minimal side effects, high patient satisfaction rates, and faster recovery times compared to either drug alone[4][5][8]. The combination is not marketed as a fixed-dose product but rather administered concomitantly by clinicians.
