Overview

Fentanyl Lauriad is an investigational mu-opioid receptor agonist formulation of fentanyl designed as a **bioadhesive buccal tablet (Lauriads technology)** for local transmucosal delivery of the potent synthetic opioid analgesic fentanyl. Like other fentanyl products, it provides strong analgesia by selectively activating mu-opioid receptors in the central nervous system, leading to inhibition of adenylate cyclase, reduced cAMP, decreased presynaptic calcium influx, and hyperpolarization of neurons, which suppresses nociceptive signal transmission.[1][9] This Lauriad formulation was developed to deliver fentanyl across the buccal mucosa for severe pain (notably cancer-related breakthrough pain), leveraging sustained local residence of the tablet on the mucosa to control systemic exposure and onset while maintaining the established pharmacology and risk profile of fentanyl, including high potency, risk of respiratory depression, tolerance, dependence, and abuse.[1][2][9]
