Overview

FEP633 and FEP600 are preclinical-stage small molecule peptide mimetics (PepMetics) developed by PRISM BioLab for the treatment of urothelial carcinoma. FEP600 functions as a mimetic of the eukaryotic translation initiation factor 4E-binding protein 1 (4E-BP1), which competitively inhibits the protein-protein interaction (PPI) between eukaryotic translation initiation factor 4E (eIF4E) and eukaryotic translation initiation factor 4G1 (eIF4G1). By disrupting the formation of the eIF4F translation initiation complex, these compounds suppress the translation of oncogenic mRNAs involved in tumor growth and progression. FEP633 is a prodrug of FEP600, currently in the final stages of lead optimization. Preclinical studies in patient-derived xenograft (PDX) models of muscle-invasive and metastatic urothelial carcinoma have demonstrated that FEP600 possesses superior anti-tumor activity compared to standard-of-care cisplatin.
