Drug intelligence / Profile preview
Ferristatin II (also known as NSC 8679 or Chlorazol Black E) is a small molecule iron transport inhibitor and polysulfonated azo dye. It acts by promoting the internalization and degradation of transferrin receptor 1 (TfR1) through a nystatin-sensitive lipid raft pathway, thereby reducing cellular iron uptake. Additionally, ferristatin II has been shown to induce the internalization of divalent metal transporter 1 (DMT1) at the plasma membrane and upregulate hepatic hepcidin expression. Due to its ability to inhibit iron uptake and suppress ferroptosis, ferristatin II is widely used as a research tool to study iron homeostasis, ferroptosis, and their roles in various pathological conditions such as traumatic brain injury (TBI), viral replication (including SARS-CoV-2), and allergic asthma.
