Overview

Ferrous iron-modified intravenous immunoglobulin (Fe(II)-IVIG) is an experimentally modified formulation of intravenous immunoglobulin (IVIG) produced by exposing standard IVIG to ferrous ions (Fe(II)). This mild modification process induces a shift in the repertoire of antibody specificities, a phenomenon referred to as leveraged polyspecificity. Unlike other modifications such as heme or low pH exposure, Fe(II)-IVIG specifically demonstrates enhanced pro-apoptotic effects on human neutrophil granulocytes. This therapeutic potential is mediated through a FAS-dependent (CD95) mechanism, which triggers programmed cell death in neutrophils. Research has shown that Fe(II)-IVIG can effectively target neutrophils primed by inflammatory mediators, joint fluid from rheumatoid arthritis patients, or neutrophils isolated from patients with acute Crohn's disease, suggesting its potential utility in treating severe inflammatory and autoimmune disorders.
