Drug intelligence / Profile preview
Fetrastobart vedotin (also known as Fetra-V, PF-08046054, or SGN-PDL1V) is an investigational antibody-drug conjugate (ADC) targeting programmed death-ligand 1 (PD-L1). Originally developed by Seagen and subsequently advanced by Pfizer following its acquisition of Seagen, the drug consists of a humanized anti-PD-L1 monoclonal antibody (fetrastobart) conjugated to the microtubule-disrupting cytotoxic payload monomethyl auristatin E (MMAE) via a protease-cleavable maleimidocaproyl-valine-citrulline (mc-vc) linker. Fetrastobart vedotin is designed to selectively bind to PD-L1-expressing cancer cells, internalize, and release MMAE to induce direct tumor cell death, bystander killing, and immunogenic cell death. It is primarily being evaluated in Phase 3 clinical trials for the treatment of previously treated PD-L1-positive non-small cell lung cancer (NSCLC) and is also under investigation for other advanced solid tumors.
