Fevipiprant is a selective, orally available small molecule antagonist of the prostaglandin D2 receptor 2 (DP2 or CRTh2), developed by Novartis. It was investigated primarily for the treatment of asthma, as well as atopic dermatitis and allergic rhinitis. Fevipiprant works by blocking the DP2/CRTh2 receptor, which plays a key role in mediating type 2 inflammation through recruitment and activation of T helper 2 (TH2) cells and other immune cells involved in allergic responses. By inhibiting this pathway, fevipiprant aimed to reduce airway inflammation and improve lung function in patients with moderate-to-severe asthma. Despite initial promise in early-phase trials showing improvements in lung function and reductions in airway eosinophilia, phase III clinical trials failed to demonstrate significant clinical benefit over placebo. As a result, development for all indications was discontinued[1][3][4][6][7].
