Overview

Fezolinetant is a nonhormonal, small-molecule, orally active, selective neurokinin 3 (NK3) receptor antagonist used to treat moderate to severe vasomotor symptoms (hot flashes) due to menopause. Its mechanism of action involves blocking the binding of neurokinin B (NKB) on the kisspeptin/neurokinin B/dynorphin (KNDy) neurons in the hypothalamus, thereby modulating neuronal activity in the thermoregulatory center and reducing hot flashes. Fezolinetant does not act as a hormone and has minimal impact on sex hormone levels. It was developed by Astellas Pharma after acquiring it from Ogeda (formerly Euroscreen). Fezolinetant is approved for use in the United States and European Union as of 2023[1][4][5][6][8].
