Overview

FG-3175 is a monoclonal antibody targeting C-C motif chemokine receptor 8 (CCR8), a G protein-coupled receptor highly and specifically expressed on immunosuppressive tumor-infiltrating regulatory T cells (Tregs) across various solid tumors. The drug is designed to deplete CCR8-positive Tregs via antibody-dependent cellular cytotoxicity (ADCC) within the tumor microenvironment, thereby enhancing anti-tumor T cell responses and potentially improving clinical outcomes in cancers such as breast, colorectal, ovarian, and melanoma. In addition to its ADCC activity, FG-3175 may also disrupt CCR8 signaling directly, which could further inhibit Treg proliferation and migration into tumors. FG-3175 is being developed by FibroGen for the treatment of solid tumors with high infiltration of CCR8-positive Tregs. As of June 2024, an Investigational New Drug (IND) application submission is planned for 2025[1][4][5].
