Overview

FG-4539 is a small molecule hypoxia-inducible factor prolyl hydroxylase (HIF-PH) inhibitor developed by FibroGen. It functions by inhibiting the HIF prolyl hydroxylase enzymes, which are responsible for the degradation of hypoxia-inducible factors (HIF) under normoxic conditions. By stabilizing HIF-1α and HIF-2α, FG-4539 activates a coordinated cellular protective response against hypoxia and ischemia. Preclinical studies have investigated its potential in treating ischemic stroke, where it has demonstrated neuroprotective effects and the ability to reduce brain tissue damage. While it shares a mechanism of action with the approved drug roxadustat (FG-4592), FG-4539 remains a research compound primarily used in experimental models of metabolic and neurological disorders.
