FG3622 is an orally bioavailable small molecule inhibitor of hypoxia-inducible factor prolyl hydroxylase (HIF-PH), developed by FibroGen for the treatment of inflammatory conditions. By inhibiting the PHD enzymes responsible for the oxygen-dependent degradation of HIF-alpha, FG3622 stabilizes HIF transcription factors, thereby activating a broad range of genes involved in the adaptive response to hypoxia. In the context of inflammatory bowel disease (IBD), this mechanism is intended to enhance the intestinal mucosal barrier, promote epithelial cell survival, and modulate local immune responses to facilitate tissue repair. While structurally related to erythropoiesis-stimulating HIF-PH inhibitors like roxadustat, FG3622 was specifically optimized and investigated for its potential to provide therapeutic benefit in gastrointestinal inflammation.
