Overview

FGF-5 peptide vaccine is an experimental therapeutic cancer vaccine designed to induce a cytotoxic T-lymphocyte (CTL) response against tumor cells overexpressing fibroblast growth factor-5 (FGF-5). The vaccine consists of synthetic peptides derived from the non-mutated FGF-5 protein, which functions as a tumor-associated antigen (TAA). Specifically, the vaccine incorporates HLA-A2 and HLA-A3 restricted epitopes—including a unique spliced peptide epitope—to ensure broad patient eligibility based on HLA haplotype. Typically formulated with the adjuvant Montanide ISA-51, the vaccine aims to stimulate the immune system to recognize and eliminate malignant cells, particularly in renal cell carcinoma (RCC) where FGF-5 is overexpressed in approximately 60% of cases. Developed by the National Cancer Institute (NCI), the drug underwent Phase 2 clinical evaluation for metastatic renal cell carcinoma, though primary clinical development has since been discontinued.
