Overview

FGF antibody refers to a portfolio of monoclonal antibodies designed to target various members of the fibroblast growth factor (FGF) family, developed by Galaxy Biotech and licensed to Roche. The lead candidate in this program, known as RG7212 (or GB-001), is a humanized monoclonal antibody that specifically binds to and neutralizes Fibroblast Growth Factor 2 (FGF2), also known as basic fibroblast growth factor (bFGF). FGF2 is a potent mitogen and pro-angiogenic factor that promotes tumor growth, survival, and vascularization by activating fibroblast growth factor receptors (FGFRs). By sequestering extracellular FGF2, RG7212 inhibits downstream oncogenic signaling pathways such as the RAS/MAPK and PI3K/AKT cascades. Another candidate in the portfolio, RG7221 (or GB-004), targets Fibroblast Growth Factor 19 (FGF19), which is implicated in the pathogenesis of hepatocellular carcinoma. Both RG7212 and RG7221 were evaluated in Phase 1 clinical trials for patients with advanced solid tumors to assess safety and preliminary efficacy. The program originated from Galaxy Biotech, an incubator company co-founded at Panorama Research, and was subsequently licensed to Roche for clinical development in oncology.
