Overview

An FGF2-neutralizing antibody is a monoclonal antibody designed to bind to and neutralize fibroblast growth factor 2 (FGF2, also known as basic fibroblast growth factor or bFGF). FGF2 is a potent mitogen and angiogenic factor that plays a critical role in tumor growth, metastasis, and drug resistance by binding to and activating fibroblast growth factor receptors (FGFRs, particularly FGFR1). By blocking the interaction of FGF2 with its receptors, these antibodies inhibit downstream signaling pathways such as MAPK/ERK and PI3K/Akt, thereby suppressing tumor cell proliferation, migration, invasion, and angiogenesis. While several anti-FGF2 antibodies (such as GAL-F2 and 3F12E7) have been developed and evaluated in preclinical models of cancer (including hepatocellular carcinoma, breast cancer, and melanoma), none have successfully progressed through clinical development.
