Drug intelligence / Profile preview
An FGF23 blocking antibody is a therapeutic monoclonal antibody designed to bind to and neutralize fibroblast growth factor 23 (FGF23), a bone-derived hormone that regulates phosphate and vitamin D homeostasis. By blocking FGF23, these antibodies restore renal phosphate reabsorption and increase the production of active vitamin D (1,25-dihydroxyvitamin D), thereby correcting hypophosphatemia and improving bone mineralization. This class of therapy is primarily developed for FGF23-excess disorders, such as X-linked hypophosphatemia (XLH) and tumor-induced osteomalacia (TIO). Burosumab (Crysvita) is the first approved drug in this class.
