Drug intelligence / Profile preview
R3-Altibody (FGFR3xFGFR3 bispecific antibody) is a preclinical-stage tetravalent, biparatopic bispecific antibody developed by Regeneron Pharmaceuticals that targets two distinct epitopes on fibroblast growth factor receptor 3 (FGFR3). By binding to epitopes located in the immunoglobulin-like domains D1 and D2/3 of FGFR3, the antibody potently blocks receptor dimerization, inhibiting receptor autophosphorylation and downstream signaling pathways such as MAPK. R3-Altibody is designed to overcome the limitations of conventional ligand-blocking antibodies and pan-FGFR tyrosine kinase inhibitors (TKIs). It has demonstrated robust efficacy in preclinical models of FGFR3-driven tumors, including muscle-invasive bladder cancer (MIBC) and lung squamous cell carcinoma, harboring common oncogenic point mutations (e.g., S249C), FGFR3-TACC3 fusions, and TKI-resistant gatekeeper mutations (e.g., V557M/L).
