FGFR4-ADC

FGFR4-ADC is a preclinical antibody-drug conjugate (ADC) being developed by Manhattan BioSolutions to target Fibroblast Growth Factor Receptor 4 (FGFR4). The drug candidate is built upon the company's specialized ADC platform, which incorporates a legumain-activated linker system designed to release its cytotoxic payload specifically within the tumor microenvironment where legumain is overexpressed. The payload typically consists of a topoisomerase I inhibitor (TOP1i), such as an exatecan derivative, or a taxoid-based agent. FGFR4 is a relevant target in several solid tumors, including hepatocellular carcinoma and certain pediatric cancers like rhabdomyosarcoma. The development of this ADC for pediatric indications is supported through a collaboration with the National Cancer Institute's (NCI) Pediatric Oncology Program (PIVOT).
