Overview

FGFR4-CAR T cells are a form of adoptive cell therapy in which a patient’s own T lymphocytes are genetically engineered to express a chimeric antigen receptor (CAR) that specifically targets fibroblast growth factor receptor 4 (FGFR4), a cell surface tyrosine kinase highly expressed in rhabdomyosarcoma (RMS) and minimally present in healthy tissues. The most studied construct is based on the anti-human FGFR4-specific murine monoclonal antibody 3A11. These modified T cells demonstrate robust cytokine production and cytotoxicity against RMS cell lines in vitro and can eliminate metastatic and orthotopic RMS tumors in vivo. Preclinical studies show high selectivity for tumor over normal tissue, with minimal off-tumor toxicity observed. The therapy is being developed primarily for pediatric patients with relapsed or refractory RMS, addressing an unmet need due to poor outcomes with conventional therapies[1][2][6].
