Drug intelligence / Profile preview
Fgl2 circular RNA lipid nanoparticle is an investigational circular RNA (circRNA) therapeutic encapsulated in lipid nanoparticles (LNPs) designed to achieve systemic overexpression of circulating fibrinogen-like protein 2 (Fgl2) [A063]. Fgl2 is an immunomodulatory molecule secreted by CD8+ regulatory T cells that plays a key role in the establishment and maintenance of spontaneous transplant tolerance (defensive tolerance) [A063]. By delivering circRNA encoding either the soluble Fgl2 (sFgl2) fibrinogen-related domain (FRED) or the full-length membrane-bound Fgl2 (mFgl2) coding region, the therapy aims to prolong allograft survival and prevent transplant rejection [A063]. In preclinical murine models of abdominal cardiac transplantation, systemic administration of Fgl2 RNA-LNPs significantly increased circulating Fgl2 protein levels and prolonged allograft survival [A063].
