FGN849 is a small molecule cytotoxic agent belonging to the indolinobenzodiazepine pseudodimer (IGN) class. It serves as the active catabolite of the antibody-drug conjugate IMGN632 (piveximab sunirine), which targets CD123. FGN849 exerts its antineoplastic effect by binding to the DNA minor groove and inducing DNA mono-alkylation. This covalent modification of DNA triggers a cascade of events including DNA damage, cell-cycle arrest, and eventually apoptosis. Developed by ImmunoGen, FGN849 was specifically selected as a payload catabolite for its high potency against malignant cells, such as those found in acute myeloid leukemia (AML), while demonstrating a more favorable safety profile and reduced myelotoxicity compared to DNA-crosslinking IGN payloads.
