FH-EB02 is a bispecific monoclonal antibody targeting B7 homolog 3 (B7-H3) and the epidermal growth factor receptor (EGFR), developed by Shanghai Henlius Biotech for the treatment of advanced non-small cell lung cancer (NSCLC). The molecule is designed with a differential affinity strategy, featuring a high-affinity arm for B7-H3 and a lower-affinity arm for EGFR. This design aims to selectively target tumor cells that co-express both antigens, thereby maximizing therapeutic efficacy while minimizing off-tumor toxicities associated with EGFR inhibition in healthy tissues. In preclinical studies, FH-EB02 has demonstrated significant tumor growth inhibition in both cell line-derived xenograft (CDX) and patient-derived xenograft (PDX) models of NSCLC, particularly in those with high co-expression of B7-H3 and EGFR.
