FH-MagIC TCR-T cells + atezolizumab is an investigational combination immunotherapy being developed by the Fred Hutchinson Cancer Center. The regimen consists of autologous CD8+ and CD4+ T cells genetically engineered to express a high-affinity T-cell receptor (TCR) specific for the MAGE-A1 cancer-testis antigen (FH-MagIC TCR-T cells), administered in conjunction with the immune checkpoint inhibitor atezolizumab. MAGE-A1 is a protein frequently expressed on the surface of various tumor cells but absent in most normal tissues, making it a selective target for cellular therapy. Atezolizumab, a monoclonal antibody targeting Programmed Death-Ligand 1 (PD-L1), is included to prevent tumor-mediated immune suppression and enhance the persistence and anti-tumor activity of the engineered T cells. This combination is primarily being evaluated in a Phase I/II clinical trial (ATTAMAGE-A1) for patients with metastatic MAGE-A1-positive triple-negative breast cancer, urothelial carcinoma, and non-small cell lung cancer.
