FH-STEAP1 IL-18 CAR T cells (also known as PRO CAR-202) is an investigational, autologous, genetically engineered chimeric antigen receptor (CAR) T-cell therapy developed by Fred Hutchinson Cancer Center and licensed to PromiCell. The therapy targets Six-transmembrane epithelial antigen of the prostate 1 (STEAP1), a cell surface antigen highly expressed in metastatic castration-resistant prostate cancer (mCRPC) and Ewing sarcoma. To overcome the immunosuppressive solid tumor microenvironment, these CAR T cells are 'armored' to secrete interleukin-18 (IL-18), a pro-inflammatory cytokine that enhances T-cell persistence, proliferation, and anti-tumor activity. FH-STEAP1 IL-18 CAR T cells are being evaluated in a Phase 1 clinical trial (NCT07803653) for patients with mCRPC following lymphodepleting chemotherapy.
