Overview

FH2201 is a small molecule inhibitor of glutaminyl-peptide cyclotransferase-like protein (QPCTL), currently being developed by Fosun Pharma USA for the treatment of solid tumors. QPCTL is an enzyme responsible for the post-translational modification of CD47, specifically the formation of pyroglutamate on its N-terminus, which is essential for the interaction between CD47 and its receptor, signal regulatory protein alpha (SIRPα), on myeloid cells. By inhibiting QPCTL, FH2201 disrupts the "don't eat me" signal mediated by the CD47-SIRPα axis, thereby enhancing the phagocytic clearance of tumor cells by macrophages and other innate immune cells. The program is currently in the IND-enabling stage.
