FHND71 is a small molecule inhibitor of the RET (Rearranged during Transfection) receptor tyrosine kinase, currently under development by Protheragen. RET is a proto-oncogene that, when activated by mutations or chromosomal rearrangements, drives the progression of various cancers, including medullary thyroid cancer and RET fusion-positive non-small cell lung cancer. FHND71 is designed to selectively inhibit the kinase activity of RET, thereby blocking downstream signaling pathways such as RAS/MAPK and PI3K/AKT that promote tumor cell growth, survival, and metastasis. The compound is currently being evaluated in Phase I clinical trials for the treatment of patients with advanced solid tumors harboring RET alterations.
