Overview

FGF-2 (Fibroblast Growth Factor-2), also known as basic FGF, is a recombinant human protein investigated for the treatment of chronic tympanic membrane perforations. In clinical trials led by D. Bradley Welling, MD, PhD, at Harvard Medical School and Massachusetts Eye and Ear, the treatment was administered topically by saturating a gelatin sponge with FGF-2 and placing it into the perforation, followed by the application of Tisseel fibrin glue. FGF-2 functions as a potent mitogen that binds to fibroblast growth factor receptors (FGFRs), triggering cellular proliferation, migration, and neovascularization to promote wound healing and closure of the ear drum. A Phase II randomized, placebo-controlled trial funded by the Department of Defense evaluated its safety and efficacy, though it did not reach statistical significance for the primary endpoint of perforation closure.
