Fibroblast growth factor 2 (FGF2) neutralizing antibodies are a class of monoclonal antibodies designed to bind and neutralize extracellular FGF2 (also known as basic fibroblast growth factor or bFGF). FGF2 is a potent pro-angiogenic and pro-survival cytokine that plays a critical role in tumor angiogenesis, lymphangiogenesis, and tumor cell proliferation. By binding to extracellular FGF2, these neutralizing antibodies block its interaction with fibroblast growth factor receptors (FGFR1–4), thereby inhibiting downstream signaling pathways such as MAPK/ERK and PI3K-Akt. This inhibition leads to reduced tumor vascularization and suppressed tumor growth. Preclinical candidates, such as Galaxy Biotech's GAL-F2 (which was licensed to Roche), have demonstrated significant efficacy in animal models of hepatocellular carcinoma and other solid tumors, both as monotherapy and in combination with anti-VEGF therapies or chemotherapy.
