Overview
Fibroblast growth factor 5 (FGF5) is a protein belonging to the fibroblast growth factor family, recognized for its involvement in cell proliferation, differentiation, and tissue repair. It functions as a tumor-associated antigen in kidney cancer, making it a target for experimental peptide vaccines aimed at eliciting an immune response. Beyond its role in oncology, FGF5 is implicated in various physiological and pathological processes, including the regulation of ferroptosis in renal tubular epithelial cells through mitophagy induction, and interaction with Fibroblast growth factor binding protein 1 (FGFBP1) in liver regeneration. FGF5 is also a key player in the Mitogen-activated protein kinase (MAPK) signaling pathway and the Sonic Hedgehog (SHH) signaling pathway. Research indicates FGF5 as a causal risk factor for conditions such as pre-eclampsia, hypertensive disorders, stroke, and atrial fibrillation. Furthermore, it has been identified as a potential biomarker for familial hypercholesterolemia and a therapeutic target in medulloblastoma and intracranial aneurysms.
