Overview

Ficlatuzumab is a potent humanized IgG1 monoclonal antibody that targets hepatocyte growth factor (HGF). By binding specifically to HGF with high affinity and specificity, it inhibits the HGF/cMET signaling pathway which is implicated in tumor growth and metastatic progression in various cancers. This pathway also acts as an escape mechanism for resistance to epidermal growth factor receptor (EGFR) blockade. Ficlatuzumab’s inhibition of HGF/cMET downstream signaling has shown strong additive antitumor effects in preclinical and early clinical studies. It is being developed primarily for treatment of cancers such as head and neck squamous cell carcinoma (HNSCC), non-small cell lung cancer (NSCLC), acute myeloid leukemia (AML), pancreatic cancer, and ovarian cancer. The drug is administered intravenously and remains investigational without FDA approval to date. AVEO Oncology developed ficlatuzumab and continues clinical trials including a phase 3 study in combination with cetuximab for recurrent or metastatic HPV-negative head and neck squamous cell carcinoma[1][3][4][7].
