Drug intelligence / Profile preview
FIDAS-5 is a potent, orally active, and cell-permeable small molecule inhibitor of methionine adenosyltransferase 2A (MAT2A) with an IC50 of 2.1 μM. It was originally developed by researchers at the University of Kentucky. FIDAS-5 competitively inhibits S-adenosylmethionine (SAM) binding to MAT2A, leading to a reduction in intracellular SAM levels. This depletion of SAM impairs cell viability, induces cell cycle arrest (G0/G1 and G2 phase), and triggers apoptosis in various cancer models, including colorectal cancer, multiple myeloma, and acute lymphoblastic leukemia. Mechanistically, FIDAS-5-mediated MAT2A inhibition also downregulates the mTOR pathway, leading to decreased protein synthesis.
