Overview

Figitumumab is a fully human IgG2 monoclonal antibody that targets the insulin-like growth factor type 1 receptor (IGF1R). By selectively binding to IGF1R, it prevents insulin-like growth factor type 1 (IGF1) from activating the receptor and blocks subsequent autophosphorylation. This mechanism inhibits IGF1-mediated signaling pathways involved in cell proliferation and survival, conferring potential antineoplastic activity. Figitumumab was developed primarily for cancer indications such as non-small cell lung cancer (NSCLC), breast cancer, prostate cancer, sarcoma, and other solid tumors. Clinical development was discontinued after phase III trials failed to show benefit and due to increased adverse events[1][2][3][5][6].
