filaminast

Filaminast is a small molecule phosphodiesterase 4 (PDE4) inhibitor developed as an analog of rolipram. It was investigated for its potential to treat asthma and chronic obstructive pulmonary disease (COPD), acting as a selective PDE4 inhibitor with bronchorelaxant and anti-inflammatory properties. Filaminast demonstrated significant selectivity for PDE4 over other phosphodiesterases and showed in vitro and in vivo activity consistent with increased cAMP levels. However, clinical development was discontinued after Phase II trials due to a narrow therapeutic window; effective doses caused unacceptable side effects such as nausea and vomiting, which are common to the rolipram class of molecules[1][3][4].
