Filanesib is a highly selective, first-in-class small molecule inhibitor of kinesin spindle protein (KSP, also known as KIF11), a motor protein essential for mitotic spindle formation during cell division. By inhibiting KSP, filanesib disrupts the formation of bipolar spindles in dividing cells, leading to mitotic arrest and apoptosis—particularly in rapidly proliferating cancer cells. It has demonstrated preclinical and clinical activity in multiple myeloma, including synergy with other agents such as bortezomib and dexamethasone. Filanesib was developed by Array BioPharma for the treatment of multiple myeloma and other malignancies but development has been discontinued for all indications[1][2][4][5][7].
