Overview

AD-2321 + AD-2322 is a fixed-dose combination (FDC) drug regimen under development by Addpharma Inc. for the treatment of hypertension. The combination consists of fimasartan, a non-peptide angiotensin II receptor antagonist (ARB), and chlorthalidone, a thiazide-like diuretic. Fimasartan works by selectively blocking the Type-1 angiotensin II receptor (AT1), thereby inhibiting the vasoconstrictor and aldosterone-secreting effects of angiotensin II. Chlorthalidone acts by inhibiting sodium and chloride reabsorption in the distal convoluted tubule of the nephron, leading to increased excretion of sodium and water, which reduces extracellular fluid volume and blood pressure. The combination is being evaluated in Phase 1 clinical trials (e.g., NCT07312799) to assess pharmacokinetic interactions and safety in healthy adult volunteers.
