FINO2 is a small molecule peroxide compound that acts as a potent inducer of ferroptosis, a form of regulated cell death characterized by iron-dependent lipid peroxidation. Unlike other ferroptosis inducers such as erastin, which deplete glutathione (GSH) via inhibition of system xc–, FINO2 induces ferroptosis through dual mechanisms: it directly oxidizes ferrous iron and indirectly inhibits the activity of glutathione peroxidase 4 (GPX4), an enzyme critical for reducing lipid hydroperoxides. This leads to the accumulation of toxic lipid peroxides and subsequent cell death. The unique structure of FINO2—specifically its endoperoxide moiety and pendant hydroxyethyl group—is essential for its activity. It has shown selectivity for tumorigenic cells in preclinical studies and represents a distinct class among ferroptosis inducers[1][3][2].
