Firicabtagene autoleucel (CRG-022, firi-cel) is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting CD22, a B-cell surface antigen widely expressed in large B-cell lymphoma (LBCL). The product uses a fully human single-chain variable fragment (scFv) derived from the m971 monoclonal antibody and incorporates 4-1BB/CD3z intracellular signaling domains. It is designed for patients with relapsed or refractory LBCL who have previously received CD19-directed CAR T-cell therapy. The construct was originally developed at the National Cancer Institute and has been evaluated in both pediatric/young adult and adult populations with relapsed/refractory CD22+ malignancies. CRG-022 is currently being studied in phase 2 clinical trials to assess its safety, efficacy, pharmacokinetics, pharmacodynamics, and immunogenicity[1][3][5][6].
