Overview

FIS-021 is a preclinical-stage small molecule inhibitor developed by Brimstalt Therapeutics, a portfolio company of Fannin Innovation Studio. The drug targets nicotinic acid phosphoribosyltransferase (NAPRT), a key enzyme in the NAD+ salvage pathway. Many solid tumors, including those of the prostate, ovary, and pancreas, overexpress NAPRT to maintain high levels of NAD+ required for rapid proliferation and DNA repair. By inhibiting NAPRT, FIS-021 aims to selectively deplete NAD+ in these tumor cells, leading to metabolic exhaustion and cell death. The program is currently undergoing in vivo evaluation for the treatment of various solid tumors that exhibit dependency on the NAPRT-mediated salvage pathway.
