fisogatinib

Fisogatinib is an orally bioavailable, potent, and highly selective small molecule inhibitor of fibroblast growth factor receptor 4 (FGFR4). It was discovered by Blueprint Medicines and is being developed in collaboration with CStone Pharmaceuticals. Fisogatinib acts by specifically binding to FGFR4 and blocking the interaction with its ligand FGF19, thereby inhibiting FGFR4-mediated signaling pathways that drive tumor cell proliferation, differentiation, angiogenesis, and survival. This mechanism targets cancers—particularly hepatocellular carcinoma (HCC)—with aberrant activation of the FGF19/FGFR4 pathway. Clinical studies have shown that fisogatinib elicits responses in patients with advanced HCC whose tumors overexpress FGF19[1][3][5][7][8].
